Barcelona’s Metro (subway) system covers the city pretty well, and will probably be the mehod of public transport you use most.
It operates from 5am to midnight (to 2am Fri–Sat nights).
It uses the same tickets and passes as the rest of the public transport system.
The central Plaça de Catalunya is one of the main Metro junctions.
Every sight, hotel, and restaurant on this site will indicate which Metro stop(s) is the most convienent.
Line L3 is the most handy. It runs from the Sants-Estació train station down Av. Paral-lel to the port, then turns to run thorugh the tourist heart of town: up La Rambla to Plaça de Catalunya and then contining along Passeig de Gracia.
Line L5 runs from the Sants-Estació train station to Diagonal, where Passeig de Gracia and the Av. Diagonal cross.
Line L4 follows Via Laietana, bordering the eastern edge of the Ciutat Vella (old city).
You can walk from most Metro stations to wherever you’re headed, but occasionally you may find that taking a bus is easier.

The Barcelona Card gets you unlimited rides on all forms of city transport (Metro, bus, funicular, etc.), plus free entry to 20 sights, and discounts (usually 20%) at most other sights, plus on activites, shops, and shows.

If you're just passing through, I'd give Barcelona at least two full days.
It is a lovely place to relax, however, so you might be happier spending 3-4 days.
